What's the difference between source and target based data deduplication?

Source based data de-duplication takes place on the client where the data is located. Typically soruce based solutions leverage new software specifically designed to perform this function at the client or device. Target based data de-duplication takes place once the data hits the target device. Target data de-duplication typically leverages your existing data protection solution that pushes data from the device you are protecting to the device that is doing the de-duplication.

My understanding is that source based data deduplication is a better choice if I want to push data over the network (e.g. remote backup). It sounds as though target-based de-dupe fits better into an existing backup methodology (e.g. VTL). Is that the right way to think about this issue?

The choice of source vs target based deduplication depends on your use model. In general, the primary use of source-based deduplication is for remote site backup although it can be used in the datacenter. Typically, source-based deduplication essentially replaces your backup application and provides centralized management.
Target based dedupe can be used to replace tape as you mention, but can also be used at remote sites. In the remote site model, you need to have a backup application and deduplication device at every remote site. The deduplication device will replicate back to a device in the datacenter. In this model, you would have separate backup application instances and devices to manage at each remote site.
